Haplotype analysis has been served for the study of molecular evolution by means of comparing of amino acid, at first and later, DNA sequences. This technique has allowed construction of evolutionary ...trees between many species. Recently, genetic distance analysis based on haplotype of RFLPs (restriction fragment length polymorphisms) in various kind of genes indicates a major division of human populations. This paper describes the data on the distribution of haplotypes associated with β globin and prion related protein (PrP) gene in Nepalese. Using 3 common RFLPs in β globin gene cluster (Hin c II 5' to ε ; Hin d III in Gγ and Aγ ) to determine linkage of sites, 3 haplotypes have been observed in Nepalese. Among 30 chromosomes, 12 have a haplotype of (+--), 9 (-+-) and 9 (---). (Table 1) As for the PrP gene, we used 2 DNA polymorphisms. (valine (V) for methionine (M) and leucine (L) for praline (P) substitution) In 16 chromosomes (Nepalese), 2 have "V" type and 14 have "M" type and none has "L" type. (Table 2) Although the sample number is not enough to lead the definite conclusion, our data provides the suggestion that the Nepalese is different from Caucasoid group, but it situates near Caucasoid rather than other Asian group population (Thai and Japanese) in dendogram of human race.続きを見る
