Abstract We have developed a liquid-phase magnetic immunoassay using SQUID. In this method, the antibodies, which specifically bind with antigen, were labeled with magnetic marker. In order to generate the sig...nal from magnetic markers, it is necessary to apply strong external field. However, it causes the aggregation of magnetic markers, which results in the noise of the free markers. In order to solve this problem, we investigated a new method for magnetic field application. In the new method, a strong field was applied for a short time for excitation of markers, while a weaker field was applied during the measurement. It was shown that this two-step application of the external field prevents aggregation of magnetic markers and effectively reduces the noise from the free markers. We demonstrated the detection of 4.7-11.9 femto-mol/l of biotin with this method.show more
