概要: 蛋白質工学によりニワトリリゾチ-ムの機能向上を目的に以下の研究を行った。1.開発済みの大腸菌によるニワトリリゾチ-ムの生産系を用いて、N末端にMetが付加し、さらに種々のアミノ酸残基の置換を施した変異リゾチ-ムを生産し、これらを活性構造に巻き戻した。2.この系で生産したリゾチ-ムのN末端は天然型と異なるため、これを天然型にするために、N末端にMet-Tyrが付加したリゾチ-ムから出発して試験管内でこれを切り落とす方法を確立した。3.酵母の発現分泌系の最適培養条件を確立し、それにより種々の変異リゾチ-ムを生産した。4.大腸菌、及び酵母の系を用いて生産した種々の変異リゾチ-ムの性質を検討した結果、以下の事がわかった。(1)触媒基であるGlu35とAsp52の置換体は、いずれもリゾチ-ム活性がほぼ消失した。(2)Asp52の置換体は、基質結合能が低下した。(3)35位に導入した解離性残基のpkはすべて異常となり、この部位に電荷をおくことは不都合であることがわかった。(4)Trp108をGlnで置換すると、活性は消失し、安定性も極めて低下した。(5)β-シ-ト上のAsn46をAspで置換すると、活性が約1/4に低下した。(6)37位および101位をGlyで置換すると、溶菌活性が上昇した。(7)14位と15位を共に欠失させた変異リゾチ-ムの熱安定性は低下したが、活性は向上した。(8)α-ラクトアルフミン様のカルシウム結合部位を構築したリゾチ-ムの還元状態からの巻戻しはカルシウム濃度で制御できた。5.一方、安定性及び活性の向上したリリゾチ-ムを生産するためには、どの様な変異を導入すればよいかを天然の変異に学ぶために、猪、豚、スナメリ(小型鯨)、犬及び、シロカン(鳥類)のリゾチ-ムを単離し、それらの一次構造と活性及び安定性を調べた。機能の向上に寄与していると考えられるアミノ酸置換を検索中である。 続きを見る
